Is jolly the best nature for Jumpluff? if so, include a moveset. and ev's please.

Indeed, Jolly is the way. The EVs vary, 252 in Spd is a given, and then it's your call. You can put the rest into Atk if you use U-Turn, or Sp.Def/Def. Even HP for better Substitutes. I'd go for Spd and HP.

Some variants:

Jumpluff @ Leftovers / BrightPowder (is it allowed?)
Ability: Chlorophyll
Jolly
- Substitute
- Leech Seed
- Sleep Powder
- U-Turn / Aromatherapy

Jumpluff @ Leftovers / Light Clay
Ability: Chlorophyll
Jolly
- Stun Spore
- Leech Seed
- Encore
- Reflect / Substitute

Bring it out in Sunny Day and it'll be a beast (with a maximum of 700 Speed).

I don't think so, Jumpluff has horrible (Atk and) Sp.Atk. MAYBE if Sunny Day is up, HP Fire with Sp.Atk EVs could do some damage, but nothing great.
Use it as a speedy supporter/annoyer, it's what it does best.

If you'd like to play around the most I see is a Giga Drain, which along with Leech Seed gets boosted by Big Root.
But if your intention is to use Jumpluff in competitive play, stick to those movesets I posted earlier.

If you're not into OU, experiment away! Just keep in mind that Substitute is a MUST.
