I have recently bred some egg move'd rhyhorns and I wanted to make one into an EV'd Rhyperior. The move-set is as follows and will not be changed.

    Fire Fang
    Crunch
    Earhtquake
    Dragon Rush

What would be the best route as far as nature, EVs, and held item?

First of all, there is a better moveset.

Rhyperior @ Expert Belt; Solid Rock
    Adamant; 252 Attack, 252 Defense, 6 HP
    Earthquake
    Stone Edge
    Hammer Arm
    Avalanche

Earthquake and Stone Edge are Rhyperior's best attacks, and Stone Edge has a good chance of exceeding Earthquake's power with a critical hit. Hammer Arm will lower Rhyperior's Speed, but that's not all that great to begin with, plus it's good for Avalanche. Avalanche will do double damage if your opponent's Pokémon attacks and damages Rhyperior before he uses it.

This moveset is supereffective against 13 of the 17 types, and can hit neutrally on the other five. Avalanche eliminates any need for Dragon Rush, as you can hit more than just Dragons with it. Crunch also isn't needed. It only helps against Ghost and Psychic, and you can already hit them very well with Stone Edge, Avalanche, and Earthquake.

Actually Avalanche ALWAYS goes last. Also, how did you get "hits 13 of 17 types super effectively, and the other 5 normally?" Slight math issue or typo? lol

Waker has a point. I can see you bred off of a Garchomp for those moves, but his set is further superb.

I'm not a Rhyperior Expert, but it needs Stone Edge AND Earthquake. Avalanche > Dragon Rush in use and power vs. Dragons. Fire is covered by Avalanche, Earthquake, Stone Edge, and Hammer Arm, making it a futile move. Although you may hit Forretress x4 with it and Skarm x2, Stone Edge already has more power from STAB than x2 Fire Fang on Skarm.

If you need help in breeding one, I have some that only need Avalanche to work just fine. Hammer Arm can be acquired later on by level.

Also, Choice Band and Focus Punch can go over Expert Belt and Hammer Arm, and Evs go more like 252 HP, 180 Def, 84 Attack, and the Choice Band helps to raise the attack missing from the lost EVs to make him more tankish.

CB focus punch is really fun to mess around with, believe me. And megahorn deserves a mention as it comes in handy often against the likes of bronzong.
