Its easy. Just pick one background and the sprites you are going to use. The best program for use (In my opinion) is the MS Paint. Good luck with your comics!

how do u sprites on sprite sheets seperate from other sprites?

you mean: "How to put sprites in another sprite/document?" or "How to recolor sprites?"???
