I was thinking like, two people running along the screen while the map is a straight-forward left to right road, is it possible to make it side-scroll forever?

My reccomendation would be using an end-of-map transfer (in the example maps there's one that connects the northern end of Lappet Town to the southern end of Route 1) to connect the eastern end of map A to the western end of map B, and then the eastern end of map B to the western end of map A, so that it makes an endless loop.

At least I think that's what you want to do, something like the endless stairs in Super Mario 64, right?

You culd do something like what Rot ate her cone said.
But honestly I would make a map where before you reach the end theres and event/player touch that transfers those events to the start of the map. but you make the start and the end of the map look the same.

Here I made a Video explaning

or you could do what I did for a road car chase and have auto tiles of a road which makes it look like the cars are moveing when in actule fact the objects are static.
