Not necessarily. Most of the time, James crossdresses or does half the stuff he does for no apparent reason. Like the times he's dressed up in a tutu. And the Flaming Moltres. And that random time when he did the entire motto in a southern belle gown when Ash and company discovered Salon Roquet. And the random time when he dressed as a bride. And, well, about 75% of every time he's been either in drag or questionable costumes.
And even then, there's still Jessibelle, who isn't a member of Team Rocket but did participate in implied BDSM for no other reason than she enjoys torturing James.

In other words, it's a matter of opinion. Personally, I think even Harley's crossdressing pales in comparison to James in a frilly pink tutu. Or, well, a skintight Moltres costume that, yes, is fairly revealing if you happen to be looking there.

Also, Jessie wears the pants in the relationship. James has been caught squealing like a pansy and handing out fashion tips. He's only started being slightly more masculine in the more recent seasons, but up until the beginning of the Hoenn arc, I'd say he was pretty effeminate too.
